Does the update unroot?

Yes.

Actually, that's overly simplified. If you are rooted, but completely stock, with all OEM files and stock recovery, you can apply this update and it will unroot. But if you're running a custom recovery, the update won't work at all. You'll have to wait for someone on XDA to post a pre-rooted zip.

Anyone not associated with Sprint or Samsung can only guess. My guess is that this will be the last, unless a bug-fix is needed.

My wife will be getting it on her bone stock S3 one of these days, so I'll get to try it out there. I'll post my impressions after.
