What are md5's and how do I check them?

I'm pretty new to flashing (only been at it about 2 months). I've read that the safe thing to do is to always check the md5's. From what I understand this ensures you have a good download of the correct file or something like that, right? Can someone walk me through how to check a md5 of a file that I downloaded? (Talk to me like a noob.) Also I saw someone say something about TWRP generating an md5 when you create a nandroid backup. How do I do that? How do I use it?

Root Explorer, Rootzbox, Rom Toolbox, etc all have this functionality.
On the PC, however...I love HashTab.
