I received an audio file from someone in my contacts list--a distant acquaintance whom I haven't spoken to in almost 2 years. For this reason I'm a little suspicious as to why this person would send an audio file with no note. Is it safe to listen to the file or should I worry about compromising my phone?

Delete the file without opening, or contact him to verify. If it's a virus you'll be glad.

I wouldn't delete it - it can't do any harm just sitting there - but I'd contact him and ask what it is.
