Can 2 different music players to access 2 different file directories (with no overlap)?

You want to limit 2 apps, each to a different directory? Not unless the app is written that way, to have a limit.

Or are you asking if you can run 2 music apps at the same time, each playing from a different directory? Possibly, depending on the phone and the apps - you'd have to try it.

So it does seem possible. Do you know of any music players that allow you to have custom file directories?

Most likely there are no music apps that use "custom" directories. All music apps read the phone's media database to find the music files.

Many music apps will let you select subsets of the files, by folder/album/directory being one of those options.
