Am I correct that to get the best audio using a pair of Aptx headphones is to use local flac 16/44 files or Tidal Hi-Fi (flac) as a source?

If I use YouTube Music with their files at 256 aac (YT Premium) doesn't the audio chain suffer by compound compression going from the lossy aac then to the lossy Aptx?

Or at that bit rate the compound compression doesn't matter?

Using a Pixel 3 if that matters.

    Thanks for the consideration.

Aptx is almost lossless. Flac is lossless. So unless the compression the file is using is really bad, it won't make much difference. (An aac file can be nearly perfect, or it can be pure garbage. Surprisingly, it does better at low bit rates than at high bit rates.) But yes, if you can hear how large the calluses on the violinists fingers are by the tone of the note, use flac. (Otherwise, you're not likely to hear any difference. Most people can't detect the difference and, among the few who can, most of them don't know the difference.)
