Will I get 6.0 without a sim?

Hi, bought a note 5 on launch day, but have been keeping my nexus updated via wifi, hoping marshmallow would come ota.

Will I get it ota if I have no simple in it, but active with wifi?

I didn't realize the new sim cards for my note are the tiny ones.

To answer your question: yes, as long as you have an active internet connection, you will get the update as soon as it becomes available for your device.

About your SIM problem:
I have exchanged all of my SIM cards for the tiny Nano SIM. That way, those SIM cards will fit into any GSM phone (with adapter) and I don't have to worry anymore whenever I change my device. Over the years, I have tried many different SIM card adapters. The best working one I found a few months ago on Amazon.
