The position of the volume button on the Nexus 6P makes it easy to unintentionally raise or lower the volume. Is there a way to lock the volume.

I realize I can be more careful when picking the phone up.

Welcome to Android Central! There's no native function for that. Try Volume Control.

There is also a program called Volume Locker. It will lock various volume settings, and if you try to change the volume, it pops up a window requiring that you confirm the change, otherwise it resets the volume to what it previously was.
