I have a Galaxy S 6 which I bought 2nd hand. When I attempted to update the main OS software I receive a message about my device being modified, yet when I use SuperuserSU or similar apps, they say my phone isn't rooted.
I'm having massive battery drain issues too (recently replaced the battery and charging port).
    Any advice would be appreciated.

"Modified" can mean rooted, it can also mean that the phone has a custom ROM flashed to it, or that it used to be rooted - and it won't update if any of those things are true. (While it was rooted, changes could have been made that will brick the phone [turn it into a brick] if it gets updated - even if it's no longer rooted.)

Updating usually doesn't decrease battery drain issues, it increases them.

What version of Android are you running now? (It's possible that you're on the latest version, and what you want is to reflash the last version before that one.) But we'd have to know which version you're running to begin to guess what's wrong, or what you should do.

And if you replaces the port yourself, you could have pinched a wire, replaced the port with a defective one - a few things that would cause a massive battery drain. Look at the battery statistics and see what's causing the drain. If it's not an app, it could be the port.
