HTCDev unlock and S-OFF are two wildly different things.

I know that the new software has a new bootloader that has so far been resistant to S-OFF.

I know of one person that has tried HTCDev unlock after the OTA and has not been able to get it to work; I haven't seen any reports one way or the other on if anyone else has tried it.

There are ways to get root, but I'm not convinced that they are fool-proof. If you are adventurous and comfortable with that sort of thing, check out XDA. Otherwise, just hold tight until the community develops proven root/s-off/unlock methods for the new software.
