I have bought a High capacity battery for Note 4. What I want to know is How long should I charge it for Initial time. And will there be any issues of using it ?

The same rules apply for all lithium batteries.

Charge it until it's fully charged. (If there's no indicator on the battery showing its state of charge, at least in percent, send it back - you'll be killing it in less than a year.) Then use it, without charging it, until it gets down to 10%-15%. Then charge it until it's fully charged again. 3 full charge/discharge cycles.

From then on, don't discharge it past 40% if you can help it. Fully discharging a lithium battery every time is the fastest way of killing it in a short time (short of chopping it in half or using it to power a welding setup).

About the only "issue" is that I prefer using a spare battery (or a few if you need that much time between charges). You swap batteries when the one in the phone reaches 40%. On the Note 4, that shouldn't take 60 seconds. (That's the equivalent of a 6440mAh battery at a lot less cost and the stock back still fits. Having 2 spares is the equivalent of a 9660mAh battery - and still cheaper than some 7000mAh "high capacity" batteries.)
