To get the very best sound you can you'd want to use an OTG cable and send bit-perfect output to an amp or receiver. I do this to send audio to my headphone amp.

However, yes, you can connect a 3.5mm minijack directly to the headphone output. You'll want to set the volume at ~75-80% on the phone and let the external speaker's amp do the rest.

I don't think the Nexus 4 have ability send digital audio out through USB though.
