Is there a best or easiest method to transfer music and data from old Samsung SG4A to the Nexus 6p ?

Yes, I understand USB cable connection to laptop and just manually create the transfer from old phone to new phone.

Today, using the old Samsung, whenever I connect Bluetooth in car, the music folder is automatically recognized and thus the question, does the folder on the Nexus 6p have to be in a specific location and with a specific name or should the data transfer be done in a different way ?

Many thanks.

Does your car stereo have its own Play/Pause/Skip Track buttons when it accesses your old phone's music via Bluetooth? Or have you been using Google Play Music or the stock Music app on your phone, and streaming the audio over Bluetooth (and therefore using the phone to control the music)?

On my 6P, I use Google Play Music and stream it over Bluetooth. (I can control the pause and play using my ZenWatch.)

I moved this to the 6P forum, since it wasn't quite related to Project Fi (where it was before).

I just received several new Project Fi phones from Google

Have no idea how car starts the Bluetooth music as soon as phone is connected.

I've seen that behavior before. It might be if the Music app is still cached in the background. You could try Force Stopping the Music or Google Play Music apps before entering the car, to see if that consistently prevents the music from starting.

Google play is not running and neither is any music app in the background.

Back to original question, what is the best method to move and does folder require specific naming convention ? Older phones have SD cards but our Project Fi phone do not

Music apps can detect music files in any directory that doesn't have a ".nomedia" file in it, so it technically doesn't matter where you put the music. But to keep things organized, I recommend moving the music files to the Music directory that is standard on all Android devices. Keep the conventional folder hierarchy of Artist>Album>Song.

And I find the easiest and most reliable way to move the files is to connect the phone to a computer via USB and use Windows Explorer to drag and drop the files. If you can't connect both phones at the same time, then use the computer's hard drive as the intermediary step.
