How to take good macro shots with note 5?

I was trying to take a pic of a big spider on its Web but it kept focusing on the background and not the spider. I tried tapping on just the spider but still focused on the wrong thing.

Try switching to Pro mode and change focus to macro. Maybe that will work better for what you're doing.
