Does the latest OTA kill root?

Most likely YES!

If you choose to root your device, you pretty much choose to forego vzw support and OTA updates.

(If you're rooted and this OTA is *better* than what you're running now, I'm not sure why you rooted in the first place)

Any ROMs you may be running that are based off the stock build should be updated very quickly. Otherwise you are better off without the update if you plan on keeping root/s-off.

Yes. Avoid the VZW/HTC OTA update.
