for the incredible, i am not sure. i know when i had my droid that if i was rooted without a custom rom and updated to the OTA update it would update but it would also remove root access. i do not see why there would be a difference but there could be.
with s-off, i would assume you would be able root without a problem by flashing a custom recovery but i am not too educated on the way the DInc works completely with root access. i have only had it for a month so most of my info is coming from previous droid experience.
if you do not want to take a risk of losing anything i would wait for the official update to leak and install it manually with root access but that is just me. by updating with the OTA file, it is questionable as to what will and will not work. Your phone should work if you decide to accept because you are not messing with anything internally that isn't on unrooted devices, except root access obviously. i just don't know what will happen to root access if you do update with the OTA.
