Now that Verizon has unlimited data again and I've upgraded to unlimited data, is there any reason I wouldn't be able to run my desktop which has a wireless card in it, connected to my Wi-Fi hotspot? I'm thinking I could do that and cut out my Comcast internet.

Usually, Verizon doesn't allow the device connected to the hotspot to access the internet (they can tell if it's the phone or another, connected, device). You can try it, but unless they've changed the rules (and carriers change things without any notice), the desktop will have access to the "router" (which is what a hotspot is), but no access to the internet. (Verizon won't complain if you try it - you just won't connect with the desktop.)
