I know that older batteries needed this for long-lasting battery life. I have heard conflicting answers on this recently, so I thought that I would ask. I will be getting my new Note 4 today so I don't want to do anything that might adversely affect battery life.

Naa I used mine right out of the box. Great thing about Samsung is if it break (battery) just buy a new one

So my dad asked the same question when he got his Note 4 yesterday. I told him to simply plug it in and then turn it on. When I got my Note 4 a week ago it had about 50% charge when I first unboxed and plugged it in. His started with about 70%.

What, you didn't already get a spare battery and charger deal and have it ready to plug in a fully charger battery..... :D

Just use it while charging it. No problems.

Im kinda worried about that, most gadgets today starts at 40-50%, because thats the best level for battery's longevity. If its higher than that, there is some possibility that it was charged before.

Last time I saw a gadget with an 70% out of box charge, it was dell venue pro 8, and it was a lemon, the battery was half damaged.
