I'm in the process of loading 30GB of music from iTunes to my new S4.

Using Easy Phone Sync app.

I'm loading these to my SD card. After they're loaded, how do I access them?
I'm sure I've got to point a player to their location.

The stock or really any music player should find them. I use iSyncr to sync, it copies music files into a folder on my SD card called, strangely enough, iSyncr. The player paired with iSyncr (Rocket Player) obviously knows where to look, but so does the stock music player, and I never had to do anything.

IF Jelly Bean works like good 'ol Gingerbread simply create a folder on your SD card (for instance name it Music). Copy N Paste the music files to the new folder. Thats it. Same with .mp4/.m4v video movie files. All the players I've used automatically locate the various files they can use. I have a feeling this is done when the operating system scans the SD card on boot up ... then again I'm only guessing.

Make folders on your SD card like Wilbur said. I created "Ringtones", "Notifications", "Music", etc. Then simply move what you have into the appropriate folder.

You are correct.

There are two ways Android picks up new files. When inserting a SD card, it simply scans all the files. Alternatively, if you are copying over the USB cable, it simply adds each file as it is copied (so there is no waiting like there is when inserting a new card).
