Does it matter which root method I use?

Is s-off necessary, if so is that someting that has to be achieved aside from rooting or can that be done via one option for both?

No it doesn't matter what method you used to root,and No you do not need s-off . You do need a custom recovery which i assume you have. You will need to flash the boot.img when you flash to a different rom.
