So, just for curiosity's sake... If tomorrow, after I update my wife's phone with the OTA ED01, I decide I want to flash back to stock and update my currently very not stock phone, will One Click work for the new build to reestablish my root so I can restore my apps with TiBu and recustomize my phone on the newer Froyo?

I don't see why not, but I guess you will find out for sure when you try.
